Knock-out/Knock-in mouse or rat (via CRISPR/Cas9)

GEAM is happy to provide comprehensive support for your CRISPR/Cas9 research project. We will share our extensive CRISPR/Cas9 experience to guide your project from conception to the creation of your desired genome-edited model.

GEAM has generated over 400 models over nearly 8 years, including knock-outs, SNPs, conditional knock-outs, reporters, and precise excisions.

We will help you build your desired model of any species, but we specialize in the generation of mouse, rat, and cell line CRISPR models.

Instead of 1.5-2 years to make a genome edited mouse, it now takes as little as 4 months!

Some examples of modifications GEAM has successfully created:

  • Gene knockout
  • SNP Generation
  • Precise excisions
  • Conditional allele
  • Reporter Generation
  • Tag insertion
  • Cre knock-in
  • Fees provided are typical for the vast majority of projects of these types. Please be aware that details of a specific project may necessitate an adjustment of the fee listed here.
  • Please note we cannot offer any guarantee to successfully recover your genome edited model. Before beginning work on your project, we will do our best to inform you of expectations given our experiences with similar projects. This information is only a best, empirical estimate, and not a guarantee of project performance.

Consultation

Get started on your gene editing project by emailing us (genomeediting@biotech.wisc.edu; animalmodels@biotech.wisc.edu). Whether you are an experienced user of genome editing strategies or you simply have an idea that would allow you to ideally address your experimental questions, GEAM is happy to provide ideas and suggestions on strategies and best-practices.

Design

GEAM staff has extensive experience genome-editing projects. Any one of many design pitfalls can seriously imperil a genome editing project, and we are happy to help guide you through this critical stage. We have designed numerous mouse, rat, Drosophila, and cell-line genome editing projects.

Reagent synthesis

We order reagents (gRNA, expression vectors, donor constructs, single-stranded donor molecules) from highly reputable commercial vendors. Once in-house, we generate purified, injection-grade reagents for rodent embryo microinjection.

Microinjection of embryos to produce gene-edits in mice and rats

Once the necessary reagents are ready, Kathy Krentz will schedule an embryo manipulation day. The gRNA, Cas9, ssODN, and/or plasmid DNA (if applicable) will be microinjected or electroporated into fertilized embryos. The manipulated one-cell embryos are then implanted into the pseudopregnant recipients and offspring are born 19 days (mice) or 22 days (rats) later. At weaning, animals are genotyped to detect presence of edits.  Founders can be bred within the Biotechnology Center or shipped to the investigators for breeding.

Genotyping

We provide a number of genotyping solutions most appropriate for your model of choice. We offer high-throughput cell line screening for CRISPR/Cas9 generated knock-out projects. We also regularly genotype F1 litters for mouse and rat models.

Pricing includes all aspects including design & embryo microinjection, all the way through to fully characterized F1 animals to start your breeding colony.

Pricing below is meant to provide an estimate. Actual project costs may differ from those listed here. After consultation, we can provide a more accurate estimate. Additional attempts to generate genome edited alleles may contribute to additional costs.
